Methyldopa tablets

Crush 3 methyldopa tablets and weigh out 2.4 g. Crush 2 tablets of hydralazine and weigh out 0.4 g. Add 0.45 g of lactose, and mix geometrically. Fill 10 capsules appropriately. Capsule size 1 is needed. 

Crush 5 methyldopa tablets and weigh out 2880 mg of the powder. Similarly crush two hydralazine tablets and weigh out 400 mg of the powder. Add the two powders to obtain 3280 mg of the total powder. Use capsule size 1 and fill 328 mg in each of the ten capsules. If the powder doesn t fill properly, add additional lactose and fill in capsule size 0. 

Crush 9 methyldopa tablets and weigh out 4200 mg of the powder. Similarly crush 6 HCTZ tablets and weigh out 700 mg of the powder. Add lactose and fill in size 0 capsules. 

Calcium phosphate, dibasic offers high hardness and faster dissolution profile than lactose for a-methyldopa tablets in wet granulation.

Carbidopa (4.75), a hydrazine analog of a-methyldopa, is an important DOPA decarboxylase inhibitor. It is used to protect the DOPA that is administered in large doses in Parkinson s disease (section 4.4.4) from peripheral decarboxylation. DOPA concentrations in the CNS will therefore increase without requiring the administration of extremely high, toxic doses of DOPA. The exclusive peripheral mode of action of carbidopa is due to its ionic character and inability to cross the blood-brain barrier. Because of this effect, carbidopa is co-administered with DOPA in a single tablet formulation as a first-line therapy for Parkinson s disease. Benserazide (4.76) has similar activity.
