Contrary to what is generally believed, the need for an effective control was always recognised, and indeed demanded, and there was a sort of control. However, it was patchy, very limited in its scope, erratic in its implementation and of little relevance to the drugs that were beginning, and were likely in the future, to appear in the market as a result of progress in pharmacology and medicinal chemistry. 

In the perspective of drug discovery it should be well acknowledged upfront that the primary role of chemogenomics resides in my opinion to provide starting points for future drug optimization projects which continue to rely on the classical medicinal chemistry and in vivo pharmacology-based design and selection principles. 

There are many years between the search for a hit, development of a lead, and then its launch as a drug. Thus, the inventory of currently marketed dmgs gives only an imperfect view of the synthetic challenges that medicinal chemists have to face. For this reason, this chapter ends with exemples of fluorinated moieties recently introduced in medicinal chemistry. These functional structural patterns, which are present on new drugs or on molecules at an early stage of development, will possibly have a strong impact in medicinal chemistry in the future. 

A major advantage of the natural products approach to drug discovery is that it is capable of providing complex molecules that would not be accessible by other routes. Compounds such as paclitaxel (Taxol, 8) or rapamycin (10) would never be prepared by standard "medicinal chemistry" approaches to drug discovery, even including the newer methods of combinatorial chemistry. Likewise, the new approach of combinatorial biosynthesis, although an important one, is unlikely in the near future to yield new compounds of the complexity of paclitaxel and camptothecin.
