Immunoglobulin activation with periodate

Two procedures have been described by Nakane and co-workers. In the first of these free amino groups on the peroxidase are blocked by fluorodinitrobenzene (FDNB) treatment prior to the production of active aldehyde groups by periodate oxidation. A recent modification of this method, described here, omits FDNB blocking and recommends periodate oxidation of the enzyme at low pH prior to coupling with immunoglobulin. ... 

Bovine brain gangliosides were oxidized by periodate ion and iV -2,4-dinitro-phenyl-L-2,4-diaminobutyric acid hydrazide was covalently attached to the aldehyde groups. The dinitrophenyl-labelled gangliosides were incorporated into mature thymocytes and these were incubated with rabbit anti-dinitrophenol and fluorescein-labelled anti-rabbit immunoglobulin G, causing uniform rings on the cell surface. Gangliosides may play a primary role in the lymphocyte activation process.
