Nucleotide immunogen preparation

Antibodies to nucleosides usually react with nucleosides equaUy or only slightly better than with nucleotides (Fig. 1). They also react well with polymers in which the bases are accessible, such as denatured DNA. When the periodate oxidation technique (with ring breakage and later reduction) is used for immunogen preparation, the resulting antibodies react slightly better with deoxyribonucleosides than with ribonucleosides. 

Polyclonal Antibodies Carcinogen modified DNA was prepared as described previously (6,7). Modified DNA samples used as immunogens contained 18 pmol of 4NQ0-adduct or 11.0 pmol of DMAB-adducts in 1 ug of calf thymus DNA, this is 6 4NQ0 adducts or 3.4 DMAB adducts per lO nucleotides.
