Identification of events and data analysis

Individual binding events between myosin and actin can be identified by monitoring the variance of the data. Myosin binding increases the system stiffness, as it introduces an additional route to the mechanical ground (the microscope covershp). This means that the Brownian motion is reduced. The data can be analysed to determine, amongst other things, the average size of the myosin powerstroke and the kinetics of the unbinding process.
