Hydroxyphenyllactic acid

In the hydrolysate of toxic components in the venom of bird-catching spiders (suborder Orthognatha, family Aviculariidae, subfamilies Grammostolinae and Theraphosinae), di-aminopropane, spermine, 4-hydroxyphenylpyruvic acid, 4-hydroxybenzoic acid, 4-hydroxy-phenylacetic acid, and 4-hydroxyphenyllactic acid have been identified. Separation and structure elucidation of the individual components of the original extract were not performed (35.36).

Unlike in humans, the major metabolic pathway of coumarin in rats is the 3,4-epoxidation pathway. After a 100-mg/kg bw oral dose of [3- C]coumarin, urinary 3-hydroxycoumarin, 7-hydroxycoumarin, ort/zo-hydroxyphenyllactic acid and ortho-hydroxyphenylacetic acid accounted for 1.8, 0.4, 0.8 and 20% of the dose, respectively. Various metabolites including ort/zo-hydroxyphenylacetic acid were detected in the faeces (Kaighen Williams, 1961). Other studies in vivo have confirmed that rats are poor 7-hydroxylators of coumarin, with urinary 7-hydroxycoumarin accounting for < 1% of the dose (van Sumere Teuchy, 1971 Lake et al, 1989a). 

The aeruginosins are a class of cyanobacterial peptide incorporating two characteristic structural features an N-terminal hydroxyphenyllactic acid residue and the highly unusual amino acid 2-carboxy-6-hydroxy-octahydroindole (Choi). In an infant with a serum phenylalanine concentration greater than 4 mg dl, the following have been recommended administration of 100 mg of ascorbic add 24 h prior to obtaining a repeat blood specimen, measurement of phenylalanine and tyrosine in blood, and evaluation of urine for the presence of phenylalanine, O-hydroxyphenylacetic acid, and phenylpyruvic acid, as well as tyrosine and tyrosine derivatives (p-hydroxyphenyllactic acid and p-hydroxyphenylpyruvic add) by chromatography and spot tests. Most infants will be found to have a transient defect that demands a new testing.

An alternate reaction of prephenic acid was found in extracts of coli nxutants blocked in the conversion to phenylpyruvate. The product of the alternate reaction is p-hydroxyphenyllactic acid. Presumably oxidation to ketotyrosine and transamination follow the formation of p-hydroxyphenyllactate to yield tyrosine. 

The elevated 4-hydroxyphenyl acids are represented mainly by p-hydroxyphenyllactic and p-hydroxyphenylacetic acids due to its instability, p-hydroxyphenylpyruvic acid is less regularly found (B26).
