Haptens with aldehyde or keton groups

Oxidation of vicinal glycols of haptens with NaI04and conjugation to protein Coupling of adenosine to BGase (Lauer and Erlanger, 1971) 

Dialyze against 50 mM phosphate buffer, pH 7.4, containing 10 mM MgCh, 10 mM 2-ME and 100 mM NaCl (several changes).

Dissolve 10 mM 0-(carboxymethyl)hydroxylamine (sold as (aminooxy)acetic acid or carboxymethoxyl amine) and 4 mM keto-hapten in a mixture of 200 ml ethanol 

Reduce volume by rotary film evaporation, add water to 40 ml and extract with ether.
