Glycolipids periodate-oxidized

Periodate-oxidize a liposome suspension containing glycolipid components according to Section 2 (this chapter). Adjust the concentration of total lipid to about 5mg/ml.

In the spermatozoa of the sea urchin Anthocidaris crassipina, nine sialoglycolipids were found, two of which were isolated and their structures established from the results of acid hydrolysis, methylation, and periodate oxidation.139 The major component of the glycolipid mixture is a disialo-glycolipid, NeuAc-(2- 8)-NeuAc-(2- 6)-Glc-Cer. The least-polar, minor component has the structure of a monosialoglycolipid, NeuAc-(2 - 6)-Glc-Cer. In these compounds, glucose is again the only neutral monosaccharide, and the sialic acid is attached to it at 0-6. 

The readily available [77, 78] l,6-di-0-benzoyl-3,4-0-benzylidene-2,5-0-methy-lene-D-mannitol has been converted [79] into the benzyl ether (60), which on periodate oxidation and borohydride reduction gives the methylene acetal (61) which readily gives 1 -O-benzyl-L-glycerol for use in lipid synthesis. The acyl derivative (62) has been used [80] in glycolipid synthesis by glycosidation of the hydroxyl group.

Both the reduced periodate-oxidized cerebroside and the ceramide were as active as the original cerebroside, suggesting that the sugar moiety is not important for activity. The sphingoid moiety of a cerebroside constitute an essential part of the activity and the fatty acid having a certain chain length and the 2-hydroxy fatty acids may be the functional structures of this glycolipid (80).
