Glycolipids periodate

Periodate-oxidize a liposome suspension containing glycolipid components according to Section 2 (this chapter). Adjust the concentration of total lipid to about 5mg/ml.

PAS staining (periodic acid - Schiff s reagent) colors compounds with vicinal hydroxyl groups, i.e., mainly oligosaccharide side chains in glycoproteins, glycolipids, and nucleic acids. The sensitivity and stability is much lower than the silver staining Protocol 2.4.2.S but more specific.

In the spermatozoa of the sea urchin Anthocidaris crassipina, nine sialoglycolipids were found, two of which were isolated and their structures established from the results of acid hydrolysis, methylation, and periodate oxidation.139 The major component of the glycolipid mixture is a disialo-glycolipid, NeuAc-(2- 8)-NeuAc-(2- 6)-Glc-Cer. The least-polar, minor component has the structure of a monosialoglycolipid, NeuAc-(2 - 6)-Glc-Cer. In these compounds, glucose is again the only neutral monosaccharide, and the sialic acid is attached to it at 0-6. 

Prepare a 5 mg/ml liposome construction in 20 mM sodium phosphate, 0.15 M NaCl, pH 7.4, containing, on a molar ratio basis, a mixture of PC cholesterol PG other glycolipids of 8 10 1 2. The other glycolipids that can be incorporated include phosphatidyl inositol, lactosylceramide, galactose cerebroside, or various gangliosides. Other liposome compositions may be used, for example, recipes without cholesterol, as long as a periodate-oxidizable component... 

The readily available [77, 78] l,6-di-0-benzoyl-3,4-0-benzylidene-2,5-0-methy-lene-D-mannitol has been converted [79] into the benzyl ether (60), which on periodate oxidation and borohydride reduction gives the methylene acetal (61) which readily gives 1 -O-benzyl-L-glycerol for use in lipid synthesis. The acyl derivative (62) has been used [80] in glycolipid synthesis by glycosidation of the hydroxyl group.
