Gluzincin

The goal of the experiments we report was to create new structural model complexes for gluzincins or carboxypeptidases. With [Zn (bdtbpza)Cl] (12) for the first time a tetrahedral zinc complex with a monoanionic W,W,0-tridentate using a carboxylate 0-donor was synthesized (41). Mice and humans possess a gene that encodes a membrane-bound gluzincin metalloen-dopeptidase (Chapter 8, Table 8.2). This enzyme hydrolyzes small peptides (<3 kDa) on the amino-terminal side of aspartate residues on various proteins. If this gluzincin loses its proteolytic activity due to mutation, BSP and DMP-1 fragments are absent and the osteoid matrix fails to mineralize. Gluzincins are proteins with the consensus HexxH( > 20)E, where the third zinc ligand, a glutamate, lies at least 20 residues C-terminal to the zincin motif (Table 2).
