Glutaraldehyde immobilization trypsin

Glutaraldehyde Immobilization of Trypsin. A sample of hydro-gel-p-amlnobenzenesulfonyl carbamate (0.1-0.2 g) was suspended in 5.0 mL pH 7 phosphate buffer solution in a 10 mL vial equipped with a stirring bar and Teflon lined screw cap. To the suspension was added an excess of 50% glutaraldehyde (0.5 mL), and the... [Pg.136]

Fiber-packed PMMA microchip Trypsin Covalent linking Fiberglass bimdles entrapped in PMMA chip by in situ polymerization. Class fibers coated with a layer of CTS to immobilize trypsin with glutaraldehyde. Digestion of myoglobin of was achieved in <5 s [81]... [Pg.315]

Recently, the immobilization of enzyme onto the nanoparticle has drawn considerable interest. For instance, Chen et al. prepared nanosized chitosan particles immobilized trypsin [47]. Trypsin was immobilized on linolenic acid modified chitosan using glutaraldehyde (GA) as crosslinker. The results showed that the activity of trypsin immobilized onto linolenic acid modified chitosan nanoparticles increased with increasing concentration of GA up to 0.07% (v/v) and then decreased with increasing amount of GA. The particle size increased (from 523 to 1372 nm) with the increasing concentration of GA (from 0.03 to 0.1% v/v). In addition, the thermal stability and optimum temperature of trypsin immobilized on nanoparticles were improved. [Pg.1351]

In addition, the GMA/EDMA copolymer proved to serve as a basic unit for the fabrication of highly permeable bioreactors in capillary format. Trypsin immobilization after epoxide ring opening with diethylamine and attachment of glutaraldehyde is mentioned as the probably most prominent example [64], The immobilization of trypsin was also carried out using another class of reactive monolithic methacrylate polymer, which is based on 2-vinyl-4,4-dimethylazlactone, acrylamide, and ethylene dimethacrylate [65]. In contrast to GMA/EDMA, trypsin can directly be immobilized onto this kind of monolith, as the 2-vinyl-4,4-dimethylazlactone moieties smoothly react with weak nucleophils even at room temperature. [Pg.11]

Derivatives of cellulose continue to be used as supports for new affinity chromatography media and for immobilization of enzymes. Trypsin has been immobilized on 4-aminobenzyl-cellulose. A penicillinase from Bacillus cereus has been immobilized by glutaraldehyde-mediated reaction with aminoethyl-cellulose. Tannin has been activated with cyanogen bromide and coupled with... [Pg.635]
