Gel electrophoresis equipment

Sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis (PAGE) analysis and agarose gel electrophoresis equipment. 

Because the PCR exponentially copies the target molecule or molecules, amplicon contamination in the laboratory is a serious concern. It is recommended that the mastermix is prepared in an isolated area, such as a PCR station equipped with a UV light. This work area should be exposed to UV radiation after use to destroy any DNA contaminants. The use of dedicated pipets and Altered pipet tips is also recommended. The template DNA should be prepared and added to the reaction in an area that is isolated from the mastermix preparation hood. The thermal cycling and gel electrophoresis should be conducted in a third work area and care should be taken not to introduce amplified PCR products into the mastermix or template preparation work areas. 

In the previously described electrophoretic methods, the capillary was filled with electrolytes only. Another mode of operation in capillary electrophoresis involves filling the capillary with gel or viscous polymer solutions. If desired, a column can be packed with particles and equipped with a frit.68 This mode of analysis has been favorably used for the size determination of biologically important polymers, such as DNA, proteins, and polysaccharides. The most frequently used polymers in capillary gel electrophoresis are cross-linked or linear polyacrylamide,69 cellulose derivatives,70-75 agarose,76 78 and polyethylene glycols. 

The equipment and reagents for gel electrophoresis are readily available and familiar to laboratory workers. Particularly noteworthy is the steady increase in the popularity of precast polyacrylamide gels since their introduction in the early 1990s (see Section 8.2.4). Precast gels provide researchers with off-the-shelf convenience and reproducibility and help to make gel electrophoresis and IEF commonplace laboratory practices.
