Fucosyltransferases assay

Neoglycoproteins can serve as substrates for glycosyl-transferases in the chemo-enzymatic preparation of more complex glycan structures. In addition, neoglycoproteins are useful as substrates in the assay for glycosyltrans-ferases. Examples are fS-l,4-galactosyltransferases (71, 72) and a-fucosyltransferase (73). 

Recently, however, several such assays have been developed for the study of carbohydrate-enzyme interactions. As mentioned previously, carbohydrate microarrays can be employed for inhibitor identification. Indeed, they were recently used by Wong and coworkers to identify fucosyltransferase inhibitors from a small library (85 compounds) of triazole-containing compounds [46,... 

The reason for this discrepancy is not known. The fucosyltransferase studied by both groups was assayed in a similar manner. Grimes (1970) points out that there is a fucosidase in 3T3 and SV-40-3T3 cells which interferes with the enzyme assay variations in this fucosidase may be responsible for the different results reported by the two laboratories.
