Complement Fixation Test for Drug-Induced Thrombocytopenia [Pg.216]

Most antigen-antibody complexes fix complement through the normal pathway. The technique of complement fixation has been largely applied to determine the titer of antibody (Lachman et al. 1973). The serum to be tested is first decomple-mented by heat at 56 C for 40 min and diluted in small tubes. An equal amount of platelets, in the presence of suspected antigen, is added to each tube. Guinea pig complement is then added. This complement fixation test is more sensitive than the determination of platelet agglutinins and is now widely adopted. [Pg.216]
