Egg ovalbumin

Simultaneous immunoassays for ovalbumin and anti-estradiol were performed on a six-channel microfluidic device within 60 s (see Figure 10.1). The limit of detection of anti-estradiol was determined to be 4.3-6.4 nM [1007], Later work reduced the LOD of the antibody to 310 pM, which equaled 2100 molecules. This compared with a theoretical detection limit of 125-525 pM [1008], In another report, chicken egg ovalbumin (600 nM) was determined by immunoassay on a Pyrex glass chip using Cy5-labeled anti-ovalbumin antibody (200 nM) [678].

Protein samples used to conduct a protein transfer were bovine serum albumin (BSA, 67,000 daltons), chicken egg ovalbumin (Ovalb, 45,000 daltons), and human serum cryo-precipitate. These agents were obtained from plasma containing a mixture of proteins including Fibrinogen (340,000 daltons), human serum albumin (HSA, 67,000 daltons), and immunoglobulin G (IgG, 47,000-56,000 daltons). The cryo-precipitate was diluted with 20 ml of buffer solution prior to separation. 

Proteolytic activity of BO, BO °", BP, and BP was examined by using horse heart myoglobin, bovine serum y-globulin, BSA, human serum albumin, chicken egg white lysozyme, and chicken egg ovalbumin as the substrates. Microtiter plates were coated with 1 or 10 p ell of hapten-chicken egg ovalbumin conjugates (hapten-OVA). The retronamine coryugate provided a more sensitive assay with a more concentrated coating solution (10 g/well) than the retronecine conjugate, which was used at a 1 pg/well concentration. Both concentrations were tested with each assay. The concentration which provided the more sensitive assay was selected for later experiments.
