Addition of nucleophiles to the cyano group of cyanothiadiazole under basic conditions takes place with unusual ease <88AG(E)434,94ACS372). Hydrolysis to the amide, for example, can be effected at 0°C in the presence of a catalytic amount of sodium hydroxide or basic ion-exchange resin. At reflux temperature, hydrazine and monosubstituted hydrazines convert 3,4-dicyano-l,2,5-thia-diazole into the l,2,5-thiadiazole[3,4-. The base-catalyzed addition of acetone to cyanothiadiazole forms an enamino ketone, used as a key intermediate for the synthesis of a number of heterocyclic ring systems, e.g. isothiazole, isoxazole, pyrazole, pyrimidine, and thiazole <77H(6)1985>. [Pg.370]

Pyridines and related nitrogen heterocyclic (azabenzenoid) compounds Polyfluoroaromatic nitrogen heterocyclic systems are all activated, relative to the corresponding benzenoid compounds, towards nucleophilic aromatic substitution. The magnitude of this activation is illustrated by the effects of a ring nitrogen, relative to C—F at the same position, for attack by ammonia [91] (Figure 9.32). [Pg.315]

Transition metal catalysis is playing an ever increasing role in the synthesis of heterocyclic ring systems. Grubb s ruthenium catalyst is very effective in enyne metathesis of (10) in which the azepine (11) is formed in high yield <94SLI020> and molybdenum metathesis is used in the preparation of pyrroloazepine derivatives <94TL6005>. Palladium catalysis of the cyclisation of N,N-dimethyl-2-(r-acetoxy-2 -propenyl) benzylamines yields 2,3-dihydro-l//-2-benzazepinium (12) salts (Scheme... [Pg.296]

Currently available materials with highly positive Ae are mainly based on benzo-nitrile structures (Tables 4 and 5). Introduction of heterocyclic ring systems (Structures 2.5 and2.6)or ester links(Structures2.7 and 3.5) lead to an increase of Ae compared with the corresponding non-heterocyclic and directly connected systems. Unfortunately, the effectivity regarding Ae of the benzoni-trile structures is decreased by a local antiparallel ordering of the dipole moments... [Pg.1202]

The quatemization reaction of the thiazole nitrogen has been used to evaluate the steric effect of substituents in heterocyclic compounds since thiazole and its alkyl derivatives are good models for such study. In fact, substituents in the 2- and 4-positions of the ring only interact through their steric effects (inductive and resonance effects were constant in the studied series). The thiazole ring is planar, and the geometries of the ground and transition states are identical. Finally, the 2- and 4-positions have been shown to be different (259. 260). [Pg.386]
