A different mechanism for dissolution slowing was reported for delavirdine mesylate tablets (81). In this direct compression formulation, the methane sulfonic acid from the drug was shown to react with the disintegrant croscarmellose sodium to form the free acid of croscarmellose and the poorly soluble delavirdine free base. The authors conclude that similar reactions could occur for any acid salt of a basic drug. 

Starch was the first disintegrant used in tablet manufacture and is still used, although it has largely been superseded by the so-called super disintegrants, croscarmellose sodium,...

Croscarmellose sodium is used in oral pharmaceutical formulations as a disintegrant for capsules, tablets, and granules. 

A model tablet formulation prepared by direct compression, with croscarmellose sodium as a disintegrant, showed no significant difference in drug dissolution after storage at 30°C for 14 months. ... 

The efficacy of disintegrants, such as croscarmellose sodium, may be slightly reduced in tablet formulations prepared by either the wet-granulation or direct-compression process that contain hygroscopic excipients such as sorbitol. ... 

Croscarmellose sodium is mainly used as a disintegrant in oral pharmaceutical formulations and is generally regarded as an essentially nontoxic and nonirritant material. However, oral consumption of large amounts of croscarmellose sodium may have a laxative effect, although the quantities used in solid dosage formulations are unlikely to cause such problems. 

Crosslinking through esterification is also possible without further additive, i.e. between the carboxyl and hydroxyl groups of the cellulose derivative chains. A striking example is the insoluble pharmaceutical tablet disintegrating agent croscarmellose sodium, which is produced by internal crosslinking mediated via pH and temperature control (see Sect 6).
