Dansyl fluorophore

The same group extended their work recently to the synthesis of a peptide-based device detecting Zn2+ in aqueous solutions. [441 The principle is to add a dansyl fluorophore on a Lys side chain of a synthetic Zn finger peptide. Upon complexation of Zn2+, an important conformational change occurs that brings the fluorophore into a different environment and leads to a change in the fluorescence properties. 

On the other hand, (3-CD and y-CD derivatives bearing a dansyl fluorophore and a tosyl unit (51-57) were prepared [53], The fluorescence behaviors of these fluorescent CDs are essentially the same as those of CD derivatives bearing two dansyl moieties. 

Marchelli used the copper(II) complex of histamine-functionalized P-cy-clodextrin for chiral recognition and separation of amino acids [27]. The best results were obtained for aromatic amino acids (Trp). Enantioselective sensing of amino acids by copper(II) complexes of phenylalanine-based fluorescent P-cyclodextrin has been recently published by the same author [28, 29]. The host containing a metal-binding site and a dansyl fluorophore was shown to form copper(II) complexes with fluorescence quenching. Addition of d- or L-amino acids induced a switch on of the fluorescence, which was enantioselective for Pro, Phe, and Trp. This effect was used for the determination of the optical purity of proline. 

Calix-DANS2 (compound 7.3) bearing two dansyl fluorophores shows remarkable sensitivity and selectivity to Hg + with a detection limit of 0.3 pM. 

X 10 . The quantum yield of the 1 1 complex is estimated to be only 2.5% of the fluorescence quantum yield of Calix-DANS2. The strong quenching upon mercury binding can be tentatively explained in terms of electron transfer from the excited dansyl fluorophore to the bound mercury ion.

A fluorescence enhancement was demonstrated when kynuramine, a substrate for amine oxidase, became bound to the enzyme under anaerobic conditions. Lin has reported that when actin was labelled with dansyl-aziridine at Cys-373 the probe showed a blue shift in the emission maximum consistent with a hydro-phobic environment. Furthermore the excitation spectrum indicated that an L-tryptophanyl and an L-tyrosyl residue were in the near vicinity and that they transfer energy to the dansyl fluorophore.
