Current value, depression

Hypoxia is one of the signatures of cyanide poisoning. One of its markers is that ATP declines to about 10% of the normal value. Since ATP forms the substrate for adenylate cyclase, the slow charmels are affected lowering the slow calcium current and depressing the contraction. In CN poisoning, as in ischemia, oxidative metabohsm is blocked and acidosis is enhanced. Acidosis depresses contractility and metabolism while sparing ATP supplies. 

Fig. 3 shows that the corrosion potentials of steel in dry and chloride-free BMIBF4 in the presence of MBT shift to more positive values, and that both anodic and cathodic currents are depressed by MBT. This indicates that MBT is a mixed corrosion inhibitor (that acts in both a cathodic and an anodic manner). Qualitatively similar shifts in corrosion potential and depression of corrosion currents on the both sides of corrosion potential are reported in Fig. 4-8 for steel, zinc and copper in the presence and absence of water and chlorides. 

The region of immunity [Fig. 1.15 (bottom)] illustrates how corrosion may be controlled by lowering the potential of the metal, and this zone provides the thermodynamic explanation of the important practical method of cathodic protection (Section 11.1). In the case of iron in near-neutral solutions the potential E = —0-62 V for immunity corresponds approximately with the practical criterion adopted for cathodically protecting the metal in most environments, i.e. —0-52 to —0-62V (vs. S.H.E.). It should be observed, however, that the diagram provides no information on the rate of charge transfer (the current) required to depress the potential into the region of immunity, which is the same (< —0-62 V) at all values of pH below 9-8. A number of clinical circumstances have been identified which are characterized by an often profoundly depressed rate of erythropoiesis (Table 6.7). Many, if not all, such conditions could be/are responsive to administration of exogenous EPO. The prevalence of anaemia, and the medical complications which ensue, prompts tremendous therapeutic interest in this haemopoietic growth factor. EPO has been approved for use to treat various forms of anaemia (Table 6.8). It was the first therapeutic protein produced by genetic engineering, whose annual sales value topped 1 billion. Its current annual sales value is now close to 2 billion. EPO used therapeutically is produced by recombinant means in CHO cells. 

However, in patients with renal failure there is a strange and currently unexplained observation in relation to non-renal clearance. If this is measured for some compounds it also is found to be depressed even though it is the kidney that is diseased and not the liver The picture becomes a little clearer if the same non-renal (presumed hepatic) clearance is measured again in patients after renal dialysis when the hepatic clearance has been found to have risen to control values. Recent animal experiments have demonstrated that the circulating inhibitor of hepatic cytochrome P450 may be parathyroid hormone. Parathyroidectomy of rats with chronic renal failure prevented the reduction in liver cytochrome activity (see Michaud et al., 2006).
