Complete sequence analysis based on cDNA was achieved for osteonectin and SPARC and indicated a disulfide-bonded, four-domain structure for the protein (Fig. 12). Remarkable features of the structure are a cluster of 16 glutamic acid residues at the N terminus and an EF-hand domain located between the last two cysteine residues, both regions having the potential for calcium-binding (Bolander et al., 1988 Engel et al., 1987). Studies with BM-40 demonstrated a reversible change in a helix from 30 to 20% upon removal of calcium. The data indicate the... [Pg.32]

The technical problems in determining amino acid sequences in H chains are quite different from those encountered with L chains. Because of the large size of an H chain, complete sequence analysis is a formidable problem. Also, the N-terminus in many H chains is blocked as a result the number of known N-terminal sequences of H chains is relatively small by comparison with L chains. An exception is the Vnni... [Pg.166]

Small analysis time The typical time needed for a complete sequence of a measurement in FIA is usually less than 60 s. This feature makes FIA very attractive when a large number of samples have to be measured. [Pg.344]
