Storage, columns

The columns should be stored in a neutral or slightly alkaline buffer (PBS or borate buffers are suitable) and sealed to prevent drying out. Storage at 4 °C in the presence of a preservative such as 0.02% sodium azide or 0.02% Thimerosal (Sigma) will prevent bacterial growth. It is important to protect the column from freezing as this will fracture agarose matrices.

For long-term column storage, it is recommended to replace the mobile phase with distilled water with 0.05% sodium azide. The columns can then be stored at room temperature. For shorter storage times, e.g., a long weekend, the column should be stored in the mobile phase after making sure that the end plugs of the columns have been tightened well. 

Minimizing entails reducing the hazards by using smaller quantities of hazardous substances in the reactors, distillation columns, storage vessels, and pipelines. When possible, hazardous materials should be produced and consumed in situ. This minimizes the storage and transportation of hazardous raw materials and intermediates. 

The column should not be exposed to unnecessarily large volumes of solvent. Columns can be stored in a clean dry atmosphere, but they should be sealed by a flame or taped for very short periods of time. Some columns need to be stored in an atmosphere of nitrogen to avoid serious loss of selectivity. The manufacturer s instructions should be followed for column storage.

Oxygen degrades the stationary phases.
