Metalloproteases. The metalloproteases are so called because they require both calcium and zinc ions as cofactors. All known metalloproteases are secreted as inactive zymogens. The metalloproteases can be divided into three main groups (A4) interstitial collagenases, type IV collagenases, and the stro-melysins. [Pg.144]

Interstitial collagenases catalyze the degradation of types I, II, and III collagen. Cleavage occurs at a single site, approximately of the distance from the carboxy-terminal end. The f and degradation products are unstable and are further broken down by either the same collagenases or other proteases. The initial bond cleaved is either a Gly-Leu or a Gly-Ile. [Pg.144]

Two main forms of collagenase IV are known, a 72-kDa form (MMP-2) and a 92-kDa form (MMP-9) (A4). The 72-kDa form is homologous to interstitial collagenase, especially at the amino and carboxy terminal ends. However, this... [Pg.144]

Synthetic Inhibitors of Bacterial and Mammalian Interstitial Collagenases... [Pg.271]

Collectively, the MMP are capable of degrading all of the major protein constituents of the extracellular matrix. The interstitial collagenases that are the focus of this review dissolve collagen fibrils by making a single scission across all three a. chains at a specific, sensitive locus of exposed type I, II... [Pg.278]
