Cloning codon optimization

Key words Gene synthesis. Codon optimization. Polymerase chain reaction. Cloning, Gene expression. Oligonucleotide design. Simulated annealing... [Pg.215]

Often, restriction sites are introduced into the synthetic gene sequence to allow for the assembly and cloning of small sections of the gene. In this case, however, internal restriction sites were omitted in order to make no compromises regarding the optimal codon usage. An Ndel and a BomHI site were added to the 5 and 3 ends of the synthetic gene, respectively. The Ndel site includes the ATG start codon, and the BomHI site immediately follows the TGA stop codon. [Pg.1024]
