Paclitaxel Cisplatin

The lack of data reinforces the need to conduct randomized trials in the area of carcinoma of the bladder, and given the radiosensitizing action of the taxanes, they are worthy of consideration in these protocols. The RTOG phase I/II trial looking at concurrent cisplatin, paclitaxel, and hyperfractionated radiation with selective bladder preservation and adjuvant chemotherapy is ongoing and it may serve as the basis for future randomized trials in the area (98).

In vitro studies with several tumor cell lines have shown vitamin C to enhance the cytotoxic activity of doxorubicin, cisplatin, paclitaxel, dacarbazine, 5-FU, and bleomycin. Vitamin C has also been shown to increase drug accumulation and to partially reverse vincristine resistance of human nonsmall-cell lung cancer cells.

Chemotherapy refers to drug administration with highly serious side effects, such as nausea, hand and foot rashes, mouth sores, and increased risk of infection, easy bruising, and so on. Therefore, liposomal carriers have been used in order to improve the drug s biodistribution and protect the patient from those side effects. The main anticancer drugs used to treat ovarian cancer are carboplatin and cisplatin, paclitaxel, topotecan, and lurtotecan. PEGylated liposomal doxorubicin has been approved as a regimen for patients with metastatic ovarian cancer refractory to both paclitaxel and platinum based-therapy [449],...

The toxicity of paclitaxel given with cisplatin appears to be dependent on the order of administration, with more severe myelo-suppression occurring if cisplatin is given first There does not appear to be any sequence dependent interaction for the combination of docetaxel with carboplatin or docetaxel with cisplatin. Paclitaxel may reduce the thromboc3i openia associated with carboplatin. The combination of carboplatin with paclitaxel appears to be more neurotoxic than carboplatin with docetaxel.
