IR spectroscopy is not a very sensitive analytical tool and is, therefore, not well suited to the detection of small amounts of material. If, however, intermediates have intense and well-resolved IR absorptions, the progress of their chemical transformation can be followed by IR spectroscopy [83,88,91-93], Near-infrared spectroscopy, in combination with an acousto-optic tunable filter, can be sufficiently sensitive to enable the on-bead identification of polystyrene-bound di- and tripeptides, even if the peptides have very similar structures (e.g., Leu-Ala-Gly-PS and Val-Ala-Gly-PS) or differ only in their amino acid sequence (e.g., Leu-Val-Gly-PS and Val-Leu-Gly-PS) [94]. Special resins displaying an IR and Raman barcode have been developed, which may facilitate the deconvolution of combinatorial compound libraries prepared by the mix-and-split method [48]. [Pg.11]

One other application where Nano-probe technology excels relative to conventional tube or flow NMR probe formats is in the area of heterogeneous samples. One of the early applications demonstrated for the Nano-probe was the acquisition of NMR spectral data for chemically modified polymer beads used in solid-phase-assisted peptide synthesis and related chemical transformations.21 23 When chemically modified beads are interrogated in a conventional NMR sample tube, the resin bead behaves as an insoluble material and at best very broad and poorly resolved spectra may be recorded. In contrast, when the same beads are placed in a Nano-probe and spun at several kHz at the magic angle, there is sufficient solvation of the pendant chemical moiety and the linker to resin bead nucleus to allow the modified portion to behave as if it is in pseudo solution, which allows reasonable NMR spectra to be recorded. Various factors affect the quality of the NMR data that can be obtained for the pendant molecule, which include the tether length and the solvent used for the measurement.23 There have been a diverse assortment of applications of Nano-probe applications reported in the literature that are discussed in further detail in Section 6.3. [Pg.19]

A HEMA-based anion exchanger developed by Alltech has been described as a universal stationary phase for the separation of a wide variety of anions [6,7]. This anion-exchange resin has been compared to agglomerated peUicular anion exchangers for the separation of anions by chemically suppressed 1C. The HEMA-based columns exhibit higher capacities for all anions and particularly for weakly retained anions such as fluoride and formate, which were completely resolved. The HEMA columns could be used for both isocratic and gradient techniques. [Pg.40]
