Carbon and Nitrogen Assimilation Tests

The choice as to whether to use broth or auxanograms for nitrogen and carbon utilization depends on individual preference. In terms of making media, we have found auxanograms preferable. 

Sterile filter housing and membrane Young culture of yeast on YM agar 

Prepare a lOX stock solution by dissolving 6.7 g Bacto-YNB in approx. 90 mL distilled water. 

Bring to final 100 mL volume with distilled water and filter sterilize. 

Aseptically, dispense 0.5 mL aliquots of stock YNB into previously autoclaved capped or cotton-plugged test tubes.
