Blood agar base

Dissolve 40 g blood agar base (Oxoid, Basingstoke) in 1 I of distilled water by standing in a boiling water bath. 

To 50 ml agar base (heated to melt and cooled to 45 ° C) add 5 ml horse blood... 

Blood agar plates Adjust trypticase soy agar base to pH 6.0 and then autoclave. Cool sterilized agar in a 50 °C water bath. Add defibrinated sheep blood to 5% final concentration immediately prior to pouring plates. Store at 4 °C. 

The bactericidal test in this, and the following tables, is based on that of Miles and Misra (1938). At the end of the given time, samples were withdrawn, diluted, and inoculated on a dried blood—agar plate. The plates were read after 48 hours at 37°C. Symbols — no growth +, up to 50 colonies + +, 50—150 colonies + ++, uncountable. (Albert, Gibson and Rubbo, 1953.)... 

Tg antibodies are directed against the Tg protein, a major constituent of thyroid colloid. Several different techniques have been used to detect and quantify TgAb in peripheral blood. These include passive hemagglutination, the agar gel diffusion precipitin technique, immunofluorescence of tissue sections, enzyme-linked immunosorbent assay (ELISA), radioassay techniques, and chemiluminescence-based immunometric assays.
