We have recently found that the anti-melanoma activity of DSF is Cu dependent,(5P) and several lines of evidence suggest that the active species is Cu(deDTC)2. First of all, the anti-melanoma activity of DSF is enhanced by the addition of CuCla and attenuated in the presence of large excess of chelators, such as bathocuproine disulfonic acid, BCPD. Likewise, DSF treatment leads to significant increases in intracellular Cu concentrations, which can also be inhibited by BCPD. Chemical studies have shown that in aqueous media, Cu(II) salts react with DSF to directly form Cu(deDTC)2 in almost stoichiometric yields, via oxidative deconq)osition of small amounts of disulfiram. [Pg.408]
