Bacto-Agar

LB agar add 1.5g Bacto-agar/100 mL LB broth. Sterilize by autoclaving... 

YT-Cm25 plates 2YT-Cm25 medium supplemented with 1.5% (w/v) bacto agar. 

All chemicals were of analytical grade and obtained from Sigma (St. Louis, MO) with the exception of bacto agar, yeast extract and peptone which were obtained from Merck (Darmstadt, Germany), and D-glucose, which was obtained from VWR. Solka-floc, a delignified pine pulp serving as the cellulosic substrate, was obtained from Fiber Sales Development (Urbana, OH). For the preparation of solutions and media, distilled water was used. 

A 1.25% solution of Difco Bacto-agar is made up in hot distilled water and sterilised by autoclaving. The agar sets on cooling and must be melted in a boiling water bath and cooled to 44°C before use. 

Prepare a basal layer of 7 ml of medium (Eagle s Glasgow modification containing tryptose and calf serum) containing 0.5% Difco Bacto agar in a 6 cm dish. 

The mouse L-929 fibroblast line was cultivated in Eagle s Minimal Essential Medium (MEM) plus 10% calf serum. Cells were seeded in 100-mm-diameter cell culture plates at 4 x 106 cells per plate and allowed to become established for 24 hr prior to use. After the monolayer was washed, 10 mL of an agar overlay consisting of 2% Bacto-Agar and 2 x MEM was added to each plate and allowed to solidify. 

LB agar (LB medium +15 g/L bacto agar, mix and autoclave to dissolve), and plates filled with LB agar containing 50 pg/ml. ampicillin. Ampicillin is added before plating but after cooling to 40°C. 

E. coli containing the plasmid of interest are taken from a glycerol stock (80) (see Note 4) and plated onto an M9 (minimal media) plate (M9 10 g M9 salts in 1 L autoclaved, distilled water, or 15 g M9 salts/L, of bacto agar to prepare plates) with appropriate antibiotics for selection.

Most applications of ImEl have been realized with commercial forms of agar. Difco Bacto Agar from Difco Laboratories, Detroit, Mich.,... 

To maintain the cells, they are grown on slants containing the peptone medium plus 1.5% Bacto-Agar. 

Luria-Bertani (LB) Broth, Miller 10 g/L Bacto tryptone and 5 g/L Bacto yeast extract 10 g/L NaCl. For plates add 15 g/L of Bacto-agar. Sterilize by autoclaving.
