This assay is altogether more difficult since three active ingredients are involved and several excipients interfere in the analysis, including one major excipient (methylparaben), which is not removed in the extraction process. In addition the active ingredients are bases which have a tendency to interact with any uncapped silanol groups in the stationary phase and it is essential to use a column which is deactivated with respect to the analysis of basic compounds. The three active ingredients are all at different concentrations in the formulation so that attention has to be paid to selection of a detection wavelength at which each component can be detected. The Folin-Ciocalteau Assay of Protein Concentration. The Folin-Ciocalteau assay is one of the most sensitive and most commonly used assays to determine protein concentration (sensitive to about 10 /rg/m I protein). This procedure employs two color-forming reactions to assay protein concentration photometrically. In the first reaction (a biuret reaction), compounds with two or more peptide bonds form a dark blue-purple color in the presence of alkaline copper salts. Four basic approaches are conducted for plant selection for antiviral screening assays 1- Random collection of plants followed by mass screening. 2- Ethnomedical approach. 3- Literature-based follow up of the existing leads. 4- Chemotaxonomic approach [12]. The second and third approaches are the most favored ones because of their cost-effective applicability. The selection based on folkloric use proved five times higher percentage of active leads than other approaches. The random approach usually affords more novel compounds with antiviral activity.
