Antibiotics industrial manufacture

Later in his career, Nagata s group made notable and useful synthetic contributions to the beta lactam field. This included developing an economically feasible synthetic method for the industrial manufacture of 1-oxacephems from penicillin G. These efforts led to the worldwide introduction in the 1980s of several clinically prominent and effective beta lactam antibiotics of the 1-oxacephem class including moxalactam. [Pg.145]

Chemferm is one of among several companies which apply penicillin acylase for the kinetically controlled industrial synthesis of semisynthetic antibiotics in aqueous environments (Scheme 37) [109-111]. Ampicillin (119) and amoxicillin (120) can be obtained by the enzyme-catalyzed condensation of 6-aminopenicillic acid (6-APA, 117) with the amide or ester of D-(-)-4-hydrox-yphenylglycine and D-(-)-phenylglycine, respectively. In a similar way, cephalexin (121) can be obtained by reaction of D-(-)-phenylglycine with 7-aminodesacetoxycephalosporanic acid (7-ADCA, 118). Penicillin acylase from diverse microbial strains such as E. colU Klyveromyces citrophiluy and Bacillus megabacterium was successfully applied for this transformation and was used in its immobilized form based on a gelatin carrier. The immobilization allows an easy separation from the reaction medium and the reuse of the enzyme for at least 50 cycles. Impressive characteristics of this transformation are yields >90%, a selectivity of >95%, and an optical purity of >99% ee. The industrial manufacture takes place in repetitive batch reactors at many locations worldwide with an annual production volume of 2,000 t. [Pg.299]

Methyl acetoacetate (MAA) and ethyl acetoacetate (EAA) are the most widely used esters they are found ia the pharmaceutical, agricultural, and allied industries. Both esters are used extensively as amine protecting agents ia the manufacture of antibiotics and synthetic sweeteners (Dane Salts) (147). Principal outiets for MAA are the manufacture of the organophosphoms insecticide dia2inon [33341-5] (148,149) and the uracil herbicides bromacil [31440-9] and terbacil [5902-51-2] (150,151) (see Insect conztiol technology Herbicides). [Pg.481]

The manufacture of benzylpenicillin (penieillin G, originally just penicillin ) is chosen as a model for the antibiotic production process. It is the most renowned of antibioties and is the first to have been manufactured in bulk. It is still universally prescribed and is also in demand as input material for semisynthetic antibiotics (Chapter 5). Developments associated with the penicillin fermentation process have been a significant factor in the development of modem bioteehnology. It was a further 30 years, i.e. not until the 1970s, before there were signifieant new advances in industrial fermentations. [Pg.149]

The freeze drying of antibiotics and blood serum have largely represented the beginning of industrial lyophilization. Neumann [3.34] wrote in 1952 The (freeze drying) temperature for the older, not well purified Penicillin preparations had to be kept surprisingly low. It could not exceed -25 °C or -40 °C and later on Today Peniciilin is manufactured as crystals without the need of freeze drying . [Pg.218]

In the US, the effort to reduce the incidence of antibiotic residues in the meat supply involves not only FSIS and FDA but also the farmers, their trade associations, feed manufacturers, and veterinarians. FSIS has expended considerable resources recently, investigating several antibiotic residue problems, such as sulfonamides and antibiotic residues in bob veal calves, and sulfonamides and chloramphenicol in pigs. Agency representatives advise the industry and other... [Pg.345]

Aeration is of importance in the fermentation industries. In the manufacture of baker s yeast, penicillin, and other antibiotics, an adequate air supply is required for optimum yields in certain submerged fermentation processes. [Pg.42]
