This chapter will outline the development of immunotoxins and describe the preclinical development required for advancing VB4-845, an anti-EpCAM targeting scFv linked to a truncated form of Pseudomonas exotoxin A(252-608), into the clinic. [Pg.650]

The use of nonrelevant animal species is discouraged as the results of such studies may be misleading (per ICH S6). Therefore the next option, when available, is to substitute a relevant transgenic animal model for a pharmacologically relevant species. Transgenic animal models for human EpCAM have been developed, but they either have a different tissue expression pattern from that seen in humans [41,43] or the model has not been validated [40], making them unsuitable for the evaluation of the safety of anti-EpCAM immunotherapeutics. [Pg.655]

Antiepidermal growth factor (EGF) antibody for targeting of glioblastoma (up to >60-fold enhanced, CAR-independent transduction) [10] Anti-EpCAM antibody for CAR-independent targeting of diverse carcinomas (10-fold increase in activity) [18]... [Pg.263]

Adhesion-based strategies make use of the protein markers expressed by cells in order to immobilize them to specific points within microfluidic devices. This has been identified as a robust and reliable method for POC applications. Typically an antibody with (specific) affinity for the protein marker of interest (e.g., CD4 in HIV diagnostics or EpCAM in CTC testing) is immobilized on the surface of a microfluidic device (Fig. 1). The capture area can be defined in various shapes, patterns, and locations, e.g., to optimize binding and/or detection. This approach was used by Nagrath et al. [3] to isolate CTCs using a microfluidic chip which consisted of an array of 78,000 microposts which were coated with anti-EpCAM antibodies. The posts increased the inner surface area of the chip and thus enhanced the efficiency of CTC capture. Up to 5.1 ml of whole blood, acquired from 116 patients with... [Pg.1886]

The CellSearch system from Veridex LLC is the only commercially available CTC detection platform. This system uses anti-EpCAM magnetic beads to enrich CTCs and then further inspects them through positive and negative labeling. [Pg.1889]
