Alfadolone and alfaxolone are two steroid anesthetics that were used in combination. However, the mixture has been withdrawn because of safety considerations regarding the solvent used, polyethoxylated castor oil (Cremophor EL), which can cause non-IgE-mediated anaphylactic (anaphylactoid) reactions (SED-10,189) (1). [Pg.72]

Anaphylactoid reactions can occur after the intravenous administration of Cremophor (SEDA-22, 526) when it is used as a diluent of alfadolone/afaxolone (SED-11,211) (2), ciclosporin (3), paclitaxel (4), propanidid (SED-11, 211) (5,6), teniposide (7), and vitamin K (8,9). Cardiorespiratory arrest after intravenous miconazole has been attributed to the histamine-releasing properties of Cremophor (10). [Pg.1016]

Athesin contains 9 mg of alfaxalone and 3 mg of alfadolone acetate per milliliter. Following intravenous administration, athesin produces sleep within 30 seconds and also causes respiratory depression and apnea.
