We have examined the use of AK for the regeneration of ATP on an industrial scale. We have developed a regeneration system using thermostable AK and thermostable adenylate kinase (AdK, E.C. 2.7.4.3) to regenerate ATP from ADP or AMP. Two reactions convert AMP to ATP. In the first reaction, AdK converts AMP and ATP to ADP (Equation 1). AK converts ADP to ATP in the second reaction, using acetyl phosphate (AcOP) as the phosphoryl donor (Equation 2). By combining these two reactions, it is evident that two equivalents of AcOP can convert one equivalent of AMP to ATP (Equation 3). [Pg.112]
