You may want to get yourself a copy of Organic Chemistry I For Dummies by Arthur Winter (Wiley). It has some detailed sections on the theory behind instrumentation and exactly how it works. 

Winter wheat plants cultivar Arthur 71 were harvested from field plots at various stages of maturity (Figure 1) as described below and refrigerated at 5°C or stored frozen at -20°C.
