Weight of packing

Often, organic chemists use a dimensionless amount loaded normalized to the weight of packing in the column, Lv. (Eq. (7.43)). This less rigorous definition does not take into account the number of sites available on the stationary phase for the product or for the impurities. Note that from column to column or repack to repack, the amount loaded at a constant Lf or a constant is a function of the porosity. [Pg.258]

Packing the Column. The quantity of stationary phase required is determined by the sample size. A common rule of thumb is to use a weight of packing material 30-100 times the weight of the sample to be separated. Columns are usually built with roughly a 10 1 ratio of height to diameter. In the microscale laboratory, two standard chromatographic columns are used ... [Pg.93]

It should be noted that the saturation capacity is determined for the column. Saturation capacities for other columns containing the same phase system can be calculated by multiplying this value by the ratio of the column volumes. If the weight of packing material in the column is known, the saturation capacity per gram of packing can be calculated. This can be used to compare different packing materials for a separation. [Pg.65]
