Cidofovir (Vistide) is an acyclic phosphonate cytosine analogue with activity against herpesviruses including CMV, HSV-1, HSV-2, EBV, and VZV. It also inhibits adenoviruses, papillomaviruses, polyomaviruses, and poxviruses. Activation of cidofovir requires metabolism to a diphosphate by host cellular enzymes. Because this activation does not depend upon viral enzymes, similar levels of cidofovir diphosphate are seen in infected and uninfected cells. Cidofovir diphosphate competes with deoxycytidine triphosphate (dCTP) for access to viral... [Pg.570]

Viruses other than HIV may be treated by blocking the action of nucleotide polymerases. Acyclovir (Zovirax, 6.56) and famciclovir (Famvir, 6.57) are used against various herpes viruses (Figure 6.22). Cidofovir (Vistide, 6.58) is active against cytomegaloviral retinitis in patients who are infected with HIV. Ribavirin (Rebetol, 6.59) is approved for use against hepatitis C virus. [Pg.141]

Huggins JW, Marteniz MJ, Zaucha GM, Jahrling PB, Smee D, Bray, M. 1998. Presented at the XII International Poxvirus Symposium, St Thomas, VI, June 6-10. [Pg.202]

Cidofovir, marketed under the name Vistide. is distributed in vials containing 375 mg of the drug dissolved in 5 mL of water. The manufacturer specifies that the drug should be kept at room temperature (68°F-77°F). The vial contents are first diluted with saline and then administered intravenously with a recommended dosage of 5 mg cidofovir per kilogram of body weight. [Pg.24]
