Vibrating microtome

For electron microscopy prepare sections (thickness 20 pm) on a vibration microtome while fragments are immersed in cacodylate buffer. At this stage Pickel et al. incubated the sections with 0.2% Triton X-100 for 30 min to improve specific detection. 

Currents from cells in an integral tissue can be recorded by making patches on cells from thin slices obtained by cutting the tissue with a vibrating microtome. Cells that maintain their connections with other cells can be visually identified using a non-inverted microscope. This technique is particularly appropriate for recording post-synaptic currents, evoked by stimulation of neighboring cells [40].

Additional samples included five replicates at each heating time without applying US vibrations. These were produced from both glass-filled and unfilled samples. Weld samples were also created between both the filled and unfilled samples. These samples were used for cross-sectioning and microtoming. Figure 2 shows a close-up of a typical sample being welded.
