Tyramine is derived by using 4-hydroxybenzaldehyde as the starting material. Vanillin and 0-vanillin can also be used in this synthesis to make the tyramine analogs 2-hydroxy-3-methoxy-B-phenylethylamine and 3-methoxy-4-hydroxy-B-phenylethylamine, respectively. Vanillin is less suspicious to purchase and the potency and elfects of the vanillin made drug are about the same as Tyramine. 

The following were isolated largely by chromatography and identified by spectral properties and by comparison with known bases N-formylnormacromerine, metanephrine, V-methylmetanephrine, syne-phrine, N- methyltyramine, V-methyl-4-methoxy- -phenethylamine, iV-methyl-3,4-dimethoxy-/3-phenethylamine, tyramine, hordenine, macromerine, and normacromerine (37).

Tyrosine and tyramine (9) are established firmly as the precursors of this group of alkaloids. Dopamine (3,4-dihy-droxyphenethylamine) (38) is incorporated in peyote and 5-hydroxy-3,4-dimethoxyphenylethylamine has been detected in peyote (Fig. 28.14). The presence of 3-methoxy-4-hy-droxy- and 4-hydroxy-3,5-dimethoxyphenylethylamine (39) in Trichocereuspachanoi, another mescaline-producing species, reveals another possible alternative pathway (Fig. 28.15). 

Belleau and Burba (1960) have used Streitwieser s asymmetric reduction method to establish the stereochemistry of the en-Z3rmatic decarboxylation of amino acids. Reduction of p-methoxy-phenylacetaldehyde with 1-deuteroisobornyloxy magnesium bromide yielded p-methoxyphenethyl-l-d alcohol. The alcohol was converted to i -l-ci-tyramine via the tosylate and azide. The fact that enzymatic decarboxylation of S-tyrosine in D2O also yields 12-l-(i-tyramine shows that this reaction proceeds with retention of configuration (Fig. 10).
