In the present chapter, we describe a detailed protocol for the detection of intraneuronal Ap peptides in transgenic mouse models of Alzheimer s disease expressing APP, focusing on the peroxidase-catalyzed immunoperoxidase method using paraffin-embedded sections. Potential pitfalls include tissue pretreatment and choice of primary antibodies to avoid lack of sensitivity and potential cross-reactivity with APP.
