Transformation of wheat by micro-projectile bombardment

Dehusk wheat seeds and place in 20% sodium hypochlorite for 10 min. 

Rinse at least three times with distilled water. 

Excise embiyos and place, scutellxun-uppermost on plates containing MS medium supplemented with 2 mg/litre 2.4-D. 

The protocol for bombardment differs according to the device used. Follow manufacturer s guidelines for loading the carrier disc into liie gun barrel. The manufiic-turer will supply instructions for the optimal use of each device. The protocol below refers to the Bio-Rad Helium 2000 gun, which is used in our laboratoiy.
