On Route 1, the deficient solubility of a drag candidate is overcome by physical means ranging from solubilization by cosolvents to complex pharmaceutical-technological systems. This kind of problem-solving is handled in the realm of the pharmacist during formulation development, and is discussed in this chapter. 

This, in turn, will affect the patterns of solubilization by cosolvents. Furthermore, a high concentration of solutes may invalidate the log-linear model, which presumes negligible volume fraction of solute and no solute-solute interactions. For solid solutes, solvent induced polymorphism may also bring additional changes in their solubilization profile.
