As a consequence of this unacceptable situation, the interest in model systems suitable for the construction and study of complex lipid/protein membrane architectures increased steadily over the years [7], The classical portfolio of model membranes used for biophysical and interfacial studies of lipid (bi)layers and lipid/protein composites includes Langmuir monolayers assembled at the water/air interface, (uni- and multi-lamellar) vesicles in a bulk (liposomal) dispersion, the bi-molecular lipid membrane (BLMs), and various types of solid supported membranes [8], All these have their specific advantages but also suffer from serious drawbacks. [Pg.90]

In contrast to polymersomes, there are various models of planar membranes monolayers at the water-air interface, free-standing bilayers, and solid-supported membranes. The functionality of proteins in natural membranes strongly depends on their mobility in the matrix, and this is thus an essential prerequisite for artificial membranes to mimic the dynamic environment of biomembranes in order to serve as templates for biomolecules. Therefore, the building blocks forming a bio-inspired membrane need to possess high flexibility to compensate the hydrophobic mismatch between the size of the biomolecules, and the membrane thickness. Furthermore, a variety of membrane properties (thickness, polarity, and surface charge) have to be considered for the successful insertion/attach-ment of biomolecules. Decoration of polymer membranes with biomolecules, either on their surfaces or inside the bilayers, can be achieved by various approaches, such as physical adsorption, insertion, and covalent binding. Compared to physical immobilization of biomolecules on... [Pg.242]

Various planar membrane models have been developed, either for fundamental studies or for translational applications monolayers at the air-water interface, freestanding films in solution, solid supported membranes, and membranes on a porous solid support. Planar biomimetic membranes based on amphiphilic block copolymers are important artificial systems often used to mimic natural membranes. Their advantages, compared to artificial lipid membranes, are their improved stability and the possibility of chemically tailoring their structures. The simplest model of such a planar membrane is a monolayer at the air-water interface, formed when amphiphilic molecules are spread on water. As cell membrane models, it is more common to use free-standing membranes in which both sides of the membrane are accessible to water or buffer, and thus a bilayer is formed. The disadvantage of these two membrane models is the lack of stability, which can be overcome by the development of a solid supported membrane model. Characterization of such planar membranes can be challenging and several techniques, such as AFM, quartz crystal microbalance (QCM), infrared (IR) spectroscopy, confocal laser scan microscopy (CLSM), electrophoretic mobility, surface plasmon resonance (SPR), contact angle, ellipsometry, electrochemical impedance spectroscopy (EIS), patch clamp, or X-ray electron spectroscopy (XPS) have been used to characterize their... [Pg.255]

Other methods of preparing bilayer membranes, which lead to more robust systems and longer lifetimes include filter-supported, solid-supported, and gel-supported membranes. Solid-supported membranes doped with ion-selective antibiotics were only used in conjunction with amperometric (porous Teflon membrane with nonactin for NH3) or conductivity (steel-supported membrane with valinomycin for measurement modes. Agar gel-supported bilayer membranes with incorporated valinomycin showed Nemstian responses for different alkali cations. ... [Pg.808]

A similar conclusion can be drawn ftom the work by Garcia-Celma et on specific anion and cation binding to lipid membranes investigated on a solid-supported membrane. In this study, defect-ftee lipid membranes were deposited on a gold support. The supported membrane has a surface that is similar to that of a ftee-standing lipid membrane. A solid-supported membrane (SSM) represents a model system for a lipid membrane with the additional benefit of being mechanically so stable that solutions may be rapidly exchanged at its surface. [Pg.33]
