2 Solid-phase synthesis of cysteine-containing peptides 

Protocol 3. Esterification of Fmoc-Cys(Trt)-OH onto 2-chlorotrityl chloride resin (49,53) 

Wash the 2-chlorotrityl chloride resin (typical loading 1.0 mmol/g) with CHjCtj. 

Add a solution of Fmoc-Cys(Trt)-OH 3 eq with respect to Cl groups on resin) and DIPEA (3.3 eq) in a minimal volume of CH2CI2. 

Terminate the reaction by adding MeOH (1 pl/1 pmol of starting chloride).
