Descriptor median values naturally depend on the composition and size of compound databases. Whenever source databases are changed, reduced, or extended in size, descriptor medians need to be re-calculated to ensure accurate MP analysis. Relatively small changes in median values can significantly alter partitioning results. 

Here, we consider it appropriate to mention briefly the so-called shape-based methods for flexible compound searching. Although these are not strictly seen in any relationship with the functional pharmacophore perception, the shape and size of compounds obviously influence activity and, in some cases, may be the main factors determining biological action.

Although ball milling can effectively reduce the particle size of compounds, prolonged milling may be detrimental in terms of compound crystallinity and stability. This has been illustrated in a study that examined the effect of ball mill grinding on cefixime trihydrate (Kitamura et al. 1989). Using a variety of techniques, it was shown that the crystalline solid was converted to an amorphous solid after 4 h in a ball mill. The stability of the amorphous solid was found to be less than that of the crystalline solid, and the samples were discoloured due to grinding. 

Both in this book and in medical practice, you will need to interconvert different units used for the weight and size of compounds and for their concentration in blood and other fluids. Table 1 provides definitions of some of the units used for these interconversions. 

In NF, it is also charge and sieving that are important. While in MF colloid interactions and in UF organic interactions are important, in NF it is the speciation, ion dissociation, organic-cation complexation, as well as the structure and size of compound that have a significant impact on rejection. Solute-solute interactions of decreasing size scale are determining rejection as membrane pore size decreases.
