Sirolimus retrospective study

Comparative studies In a two-center, retrospective study in 409 renal transplant recipients, the primary calcineurin-based immunosuppressive regimen was converted to an mTC)R inhibitor-based regimen at least 3 months after transplantation [37 ]. The rates of mTOR inhibitor withdrawal because of adverse reactions among those who converted to everolimus ( = 220) or sirolimus ( = 189) were compared. Everolimus was withdrawn in 69 patients (31%) and sirolimus in 43 (23%). The most important causes were severe infections (everolimus 2.3% sirolimus 4.8%), pneumonitis (everolimus 6.8% sirolimus 4.8%), acute rejection episodes (everolimus 4.1% sirolimus 1.6%), proteinuria (everolimus 4.1% sirolimus 1.6%), renal function deterioration (everolimus 2.3% sirolimus 2.1%), and severe rashes (everolimus 2.3% sirolimus 0.5%). Although the overall incidence of withdrawal because of adverse reactions was higher with everolimus, the frequency of severe adverse reactions was not higher. [Pg.615]

In a retrospective study of 548 heart transplant patients treated with sirolimus (34,3%) or everolimus (65.7%),... [Pg.595]

A retrospective study at 5 years post renal transplant in 131 patients (63 on sirolimus, 68 on cidosporin) living with a functional graft at 1 year showed the adverse events leading to discontinuation of sirolimus were mouth ulcer, proteinuria, pneumonia, peripheral oedema, hypercholesterolaemia, hyperlipidaemia and hypertriglyceridaemia [39 ]. [Pg.595]

In a retrospective study of 25 paediatric renal transplant patients on sirolimus, side effects noted in descending order (4 2%) for each were hyperlipidaemia, proteinuria, anaemia, interstitial pnemnonia, nasal acneiform lesions, lower extremity oedema and hypokalaemia [42 ]. [Pg.595]

A retrospective study of lung and heart transplant patients who received sirolimus-based immunosuppression as a second-line agent after initial therapy with calcinemin inhibitors showed nine patients with cavitatory lung disease. Some patients showed complete resolution, whereas others had... [Pg.595]

In a retrospective study, 12 renal transplant patients (16.7%) on sirolimus developed interstitial pneumonitis. [Pg.595]

Haematology In a retrospective study comparing 67 cardiac transplant recipients treated with sirolimus to 134 matched cardiac transplant recipients not on sirolimus, a higher incidence of venous thromboembolism was seen in patients treated with sirolimus (12% vs 7%, p=0.03) [46 ]. [Pg.595]

Skin A retrospective study compared 50 renal transplant patients receiving sirolimus to 50 renal transplant patients not receiving sirolimus. In the sirolimus group the most common dermatologic disorders were skin infection (78%),... [Pg.595]
