Single Channel Experiments

To prepare a-saxitoxinol, saxitoxin was reduced with sodium borohydride and purified by chromatography on IRP-64 and Bio-Gel P2 (14,22,23). Current records from single-channel experiments using this material are shown in Figure 13. Note the absence of blocking events with long dwell times that would indicate the presence of residual saxitoxin.

The proton experiment is a so-called single channel experiment the same channel is used for sample irradiation and observation of the signal, and the irradiation frequency is set (automatically) to the resonance frequency of the protons at the magnetic field strength used by the spectrometer. 

For Affymetrix (Santa Clara, CA) (see below) and for other single-channel experiments, the hybridization scheme is straightforward, but for two-channel platforms, the allocation of samples to arrays is important to prioritize the primary scientific question of the study. Furthermore, balanced designs should be used so that treatments are not confounded with technical issues such as dye assignments, batch of slides, or day of hybridization. A better understanding of the activator-veratridine interaction would require single-channel experiments which, however, are scarce. In cultured cardiomyocytes a mixture of veratridine and ATX II induces, on steady depolarization, frequent openings to a low conductance substate (y=4.6 pS). This can be seen in veratridine alone only after voltage jumps and for a short time whereas in ATX II alone a different substate (7 11.5 pS) is observed.
