Mazindol (Sanorex) produces typical amphetaminelike anorexia and CNS stimulation. The drug inhibits NE uptake in a manner identical to the tricyclic antidepressants. Its very potent appetite suppressant ability was found while being evaluated as an antidepressant. Mazindol does not contain the P-phenethylamine foundation within its frame. It is structurally the enol form of an imidazole-substituted benzophenone, probably in equilibrium with its isoindole form (Eq. 9.12) in solution. [Pg.411]

Mazindol (Sanorex ) is a phenyl-substituted imidazobenzoisoindole that inhibits both NE and dopamine reuptake transporters. It exhibits high affinity for the NET (rat pKj = 9.3), good affinity for dopamine transporter (rat pKj = 7.8), and a 5-HT NE ratio of 224. Dopamine reuptake inhibitors suppress appetite thus, mazindol is approved to be marketed as an appetite suppressant. [Pg.860]
