FRET is an extremely useful phenomenon when it comes to the analysis of molecular conformations and interactions. F or the analysis of interactions, in which two separate molecules are labeled with an appropriate pair of fluorophores, an interaction can be shown by observing FRET. Further, FRET can be used as a type of spectroscopic ruler to measure the closeness of interactions. Proteins, lipids, enzymes, DNA, and RNA can all be labeled and interactions documented. This general method can be applied not only to questions of cellular function like kinase dynamics [3] but also to disease pathways, for example, the APP-PS1 interaction that is important in Alzheimer s disease (AD) [4], Alternatively, two parts of a molecule of interest can be labeled with a donor and acceptor fluorophore. The bulk density of the feedstock at ambient temperature and pressure should be measured prior to the design of a new screw, especially if it contains in-plant recycle resin. The measurement method is extremely simple and requires only a calibrated cell and a scale. A calibrated measuring cell with a volume of 500 cm can easily be constructed by welding a thin-walled metal pipe to a flat sheet of metal, as shown in Fig. 4.2. The bulk density is measured by filling the cell with feedstock, leveling the top with a steel ruler, and then weighing the cell contents. The remaining useful life evaluation routine (RULER) is a useful monitoring program for used engine oils. This chapter is concerned with the precision of the atomic arrangement that we derive from a crystal structure analysis. Therefore it is necessary at this point to pause and discuss errors and estimated standard deviations, and the methods used to assign a confidence or weight to a measurement. Precision, which should not be confused with accuracy, is a measure of how closely a series of measurements of the same quantity agree with each other.Accuracy is a measure of how close a measurement is to the true value. If rulers have become distorted with time, measurements made with them may be highly precise but are unlikely to be accurate. The true electron density in a crystal is not yet attainable by theory or experiment. As detection and theoretical methods improve, we hope that the data on the geometry of molecular structures, measured in various ways, will converge and approach those of the true molecular structure. [Pg.390]

Whether the chart or the ruler is used to plot the poles of reflecting planes, they are indexed in the same way as back-reflection patterns. The location of the projected poles is quite different for the two x-ray methods. The poles of planes responsible for observed spots on a transmission film are all located near the edge of the projection, since such planes must necessarily be inclined at small angles to the incident beam.
