FIGURE 2 Colocalization of imported HSA-SKL with catalase and cytosol dependence of peroxisomal protein import. Cells permeabilized with 0.2 U/ml SLO were incubated with HSA-SKL and processed for double-labeling indirect immunofluorescence. Cells were incubated simultaneously with guinea pig anti-HSA (A) and rabbit anticatalase antibodies (B) followed by species-specific secondary antibodies coupled with FITC (A) and rhodamine (B). Cells were permeabilized with 2.0 U/ml SLO and incubated with HSA-SKL in the absence (C) or presence (D) of external cytosol in the form of rabbit reticulocyte lysate. Bar = 20 m.
